Product Introduction

This product combines multiple protease and phosphatase inhibitors to effectively prevent protein degradation by proteases and phosphatases during extraction and purification. It contains the classic inhibitors Bestatin, Aprotinin, Leupeptin, E-64, NaF, Sodium Orthovanadate, β-glycerophosphate, Sodium Pyrophosphate, etc., and is suitable for protein purification, immunoblotting, co-immunoprecipitation, immunofluorescence, immunohistochemistry, and kinase activity assays.

It does not contain AEBSF or similar substances, so it is compatible with mass spectrometry (MS). Compared with DMSO-based cocktails, this aqueous formulation can be stored in a refrigerator without freezing.

Instructions for Use

Standard Operating Procedure
  1. First Solubilization: Solubilise for first use in a 55 °C water-bath and vortex to homogeneity (this step is essential).
  2. Storage After Solubilization: After solubilisation the cocktail can be kept at 4 °C and used within 3 months; vortex again before each use. If re-frozen, repeat the 55 °C water-bath/vortex cycle.
  3. Dilution: Add the cocktail to the target solution at 1:100 (v/v) and mix thoroughly (e.g., 10 µL cocktail per 1 mL cell-lysis buffer).

Notes

  1. For research use only; not for clinical diagnosis or therapy.
  2. Protease and phosphatase levels vary among sample sources; the recommended routine dilution is 1:100. If higher activities are encountered, increase the cocktail concentration accordingly.
  3. This is a high-concentration solution; freeze-thaw may produce a small amount of crystalline precipitate. This is normal. Warm in a ≤55 °C water-bath to dissolve; any residual insoluble material after thorough mixing will dissolve upon dilution and will not affect performance.
  4. This product is EDTA-free; add EDTA separately if required.
  5. Long-term storage of this concentrated solution may generate trace crystalline precipitate; this is normal and does not influence experimental results.
  6. Wear a laboratory coat and disposable gloves for your safety and health.
