Cleaved Caspase 

Caspase Family: Caspases are a class of cysteine proteases and key executors of apoptosis (programmed cell death). Under normal conditions, they exist in an inactive zymogen form. When cells receive apoptotic signals, these zymogens are activated into active enzymes.

Formation of Cleaved Caspase: During apoptosis, caspase zymogens are cleaved to form active enzymes. This cleavage is irreversible and marks the irreversible stage of apoptosis.In many diseases, abnormal regulation of apoptosis is closely related to the occurrence and development of the disease. For example, in cancer, cancer cells may escape apoptosis by inhibiting the activation of Caspase; in neurodegenerative diseases, excessive apoptosis may lead to the death of nerve cells.

Types of Caspases

• Caspase-3: It is the most classic apoptotic executioner enzyme. Its cleavage fragments are commonly used as markers of cell apoptosis.

• Caspase-8: It mainly participates in the extrinsic apoptotic pathway, which is activated through death receptors (such as Fas).

• Caspase-9: It mainly participates in the intrinsic apoptotic pathway, which is activated by cytochrome C released from mitochondria.
