Strategies for Different PTMs
🔹 Phosphorylation
Immunogen: Phosphopeptides (usually 13-18 aa) with phosphorylated residue. Counter-screen with non-phosphopeptide to ensure site specificity.
🔸 Acetylation/Methylation
Modified peptides (acetyl-lysine, methyl-arginine). Critical counter-screening against unmodified and other modified forms.
🔸 Small Molecules (Haptens)
Conjugation to carrier proteins (KLH/BSA) via suitable functional groups. Screening against free hapten and carrier alone.
🔸 ADC Payloads
