Colony Stimulating Factors (CSFs)


The colony-stimulating factor (CSF) family comprises soluble glycoproteins that govern the survival, proliferation and lineage-specific differentiation of hematopoietic progenitors while activating mature blood cells. Four major members are recognized: granulocyte-macrophage CSF (GM-CSF), macrophage CSF (M-CSF/CSF-1), granulocyte CSF (G-CSF) and multipotential CSF (IL-3). GM-CSF, G-CSF and M-CSF engage distinct α-chains coupled to either the common β-subunit (GM-CSFRβ) or LIFRβ, triggering JAK2-STAT5/3, PI3K-AKT and ERK cascades that promote myeloid commitment toward granulocytes, monocytes/macrophages or dendritic cells. Beyond hematopoiesis, CSFs contribute to embryo implantation, placental development, tissue repair and tumor microenvironment remodeling. Recombinant G-CSF (filgrastim) and GM-CSF (sargramostim) are standard of care for chemotherapy-induced neutropenia, stem-cell mobilization and immune augmentation, and are stockpiled for acute radiation syndrome, leukemia and adjuvant immunotherapy.
