Dye Introduction

Fluor488 is a sulfonated rhodamine green derivative. The introduction of two sulfonate groups significantly enhances Fluor488's water solubility, pH stability, and photostability. Unlike fluorescein-based dyes, Fluor488's fluorescence remains constant over a broad pH range (4-10). The multiple negative charges prevent intermolecular aggregation, reducing fluorescence self-quenching or protein aggregation when multiple dye molecules label the same protein. Fluor488 is an excellent alternative to fluorescein (FITC or FAM), performing exceptionally well in applications such as bioconjugate single-molecule detection, fluorescence correlation spectroscopy (FCS), and fluorescence polarization (FP) assays. By all metrics, Fluor488 is one of the premier water-soluble small-molecule green fluorescent dyes available.

Labeling Principle
