TNF Family

The tumor necrosis factor superfamily (TNFSF) is a group of proteins that play a wide and important role in biology. Currently, 19 ligands have been identified in this family, such as TNF-α, TNF-β, CD40L, FasL, TRAIL, RANKL, etc., and 29 receptors correspond to them. TNFSF members are all type II transmembrane proteins, and their C - terminal contains a TNF - homologous domain, which can promote the formation of trimeric complexes of ligands and receptors, thus activating the intracellular signaling pathway. These ligands regulate a variety of cellular functions by binding to different receptors, including immune response, inflammatory response, cell proliferation, differentiation, apoptosis and embryogenesis, etc., playing a key role in maintaining body homeostasis and responding to external stimuli. For example, TNF - α is abnormally expressed in a variety of tumors, which can promote tumor growth, angiogenesis, invasion and metastasis, and at the same time inhibit apoptosis, while TRAIL can selectively induce tumor cell apoptosis and become a new target for tumor treatment. In addition, the over - expression of CD40L is related to a variety of autoimmune diseases, such as lupus erythematosus and rheumatoid arthritis, and the abnormal RANKL/RANK signal is related to osteoporosis.
